— PAGE II— 摘 要 白细胞介素-1( IL-1 ) ，是参与免疫反应和炎症过程的重要细胞因子，分子量为17 kD。 IL-1有两种分子结构:IL-1α和IL-1β。IL-1具有广泛的免疫调节作用，并有致热和介导炎症的作用，此外，对骨代谢及动物繁殖均具有重要影响，它的生物学功能是通过与相应高亲和力受体结合而介导的。一般认为IL-1主要介导非特异性免疫反应和调节免疫系统的功能。但近年来研究发现IL-1及其受体也存在于中枢神经系统。它做为一种神经递质、神经调质或生长因子参与了中枢神经系统的生理及病理活动变化。而且IL-1与其特异受体结合后，通过某些调节蛋白作用，激活胞膜上或胞浆内的多种磷脂酶，产生多种信使类物质。IL-1作用无明显的种属特异性，人IL-1可作用于小鼠源性的细胞。 本实验通过对鹅血细胞总RNA的反转录产物进行PCR扩增获得IL-1基因,用回收试剂盒将其回收并与pMD18―T载体连接，转入到E.coli JM109中，得到鹅IL-1基因的克隆，为今后进一步对鹅IL-1的基因表达、生物学活性和应用进行研究奠定了基础。 关键词: 白细胞介素-1, 转化 , 克隆 摘 要 Abstract Interleukin-1(IL-1), molecular weight 17kD, is a important cell factor that participates in immune reactions and inflammation processes. IL-1 has two molecular structures: IL-1αand IL-1β.IL-1 has comprehensive immunoregulation function, and has the function of producing heat and mediating inflammation. In addition, it has an important impact on the metabolism of bones and the propagation of animals, its function of biology mediates by combining with high appetency receptors. Generally speaking, it mediates non-specific immune reactions and regulates functions of immune system. According to recent studies, IL-1 and its receptors were also found in the central nervous system (CNS). IL-1 acting as a neurotransmitter, neuromodulator/growth factor,involves in normal physiological processes and pathogenesis of diseases in the CNS. Further more, IL-1 activates multiplicate phosphatide enzymes on cell membrane and plasm, and produces many kinds of messenger substances by the functions of some regulate proteins after it combines with its specific receptors. IL-1’s functions have no obvious differences in different genera, human IL-1 could effect on the cell that come from rat. This experiment enlarged the production of RT-PCR from total RNA in Gooses’ blood corpuscles, and got the gene of Goose IL-1. We reclaimed the production by using DNA gel extraction kit, and connected it with pMD18―T vector., transformed it to E.coli JM
